PHARMACHOLOGIC EFFECT

Anti-tuberculosis drug.
Is active against mycobacteria tuberculosis. The pharmacological action of phenazide is based on the modification of the isoniazid molecule by complexation with iron. This ensures greater safety of chemotherapy for tuberculosis, since the iron-blocked chelate of the molecule of hydrazine isonicotinic acid (GINC) loses its ability to interact with the active centers of metal-containing enzymes, and the inclusion of the primary amino group of hydrazine in the chelate cycle of the complex prevents interaction with N-acetyltransferase. In this regard, phenazide is a low-toxic drug, the use of which does not require the correction of single and course doses of the drug depending on the rate of its acetylation.
PHARMACOKINETICS

Suction

After ingestion, absorption is slow.
The time to reach C max in blood plasma is 5-6 h.
Excretion

T 1/2 is 7.2 hours.

INDICATIONS

- complex therapy of tuberculosis of various localization (including with concomitant diseases of the central nervous system, liver and poor tolerance of GINC drugs).

DOSING MODE

The drug is prescribed inside.
Single dose 250 mg.
To clarify the individual tolerability of the drug on the first day appoint 1 tab.
in the morning after eating. In the absence of adverse reactions on the second day and throughout the following days, the drug is prescribed at a dose of 250 mg (1 tab.) 2 times / day (morning and evening) 30-40 minutes after meals. The total duration of treatment is up to 6 months.
SIDE EFFECT

From the cardiovascular system: cardiotoxic action, acceleration of thrombogenesis processes, myocarditis are possible.

From the side of the central nervous system: a headache.

On the part of the digestive system: dyspeptic phenomena.

From the side of metabolism hemosiderosis of the parenchymal organs.

On the part of the endocrine system: hypothyroidism of the thyroid gland.

CONTRAINDICATIONS

- pulmonary heart failure;

- IHD;

- the presence of changes from the coagulation system of the blood;

- Thyroid dysfunction;

- Pregnancy;

- lactation period;

- childhood;

- Hypersensitivity to the components of the drug.

PREGNANCY AND LACTATION

The drug is contraindicated for use in pregnancy and lactation (breastfeeding).

APPLICATION FOR CHILDREN

Contraindication: children's age.

SPECIAL INSTRUCTIONS

In the course of treatment, phenazide requires systematic monitoring (at least 1 time / month) of ECG and coagulogram.

OVERDOSE

Data on the drug overdose of phenazide are not available.

DRUG INTERACTION

Since phenazid is a derivative of the GINK (chelate complex of isoniazid and ferrous iron), the combined use of phenazide and isoniazid is contraindicated.
