PHARMACHOLOGIC EFFECT
The second-line anti-tuberculosis drug is close to ethionamide by its chemical structure. It acts bacteriostatically. Between the protionamide and ethionamide there is complete cross-resistance.
PHARMACOKINETICS
After oral administration, the protionamide is well absorbed from the digestive tract. C max in blood plasma is achieved after 2 hours. It is widely distributed in tissues and body fluids, including cerebrospinal fluid. Metabolized with the formation of 2 metabolites - active sulfoxide and inactive. It is excreted in urine, less than 1% in the form of unchanged substance.
INDICATIONS
Tuberculosis (with resistance to therapy or intolerance of drugs I series).
DOSING MODE
For oral administration in adults, use in an initial dose of 250 mg 3 times / day, then - 500 mg 2 times / day. For children, the daily dose is 10-20 mg / kg.
SIDE EFFECT
On the part of the digestive system: nausea, vomiting, diarrhea, hypersalivation, metallic taste in the mouth, impaired liver function.
From the side of the central nervous system and the peripheral nervous system: sleep disturbances, arousal, depression, anxiety; rarely - dizziness, drowsiness, headache, asthenia; in isolated cases - peripheral neuropathy, optic neuritis.
From the side of the cardiovascular system: postural hypotension, tachycardia.
CONTRAINDICATIONS
Acute gastritis, peptic ulcer of stomach and duodenum, erosive-ulcerative colitis, acute hepatitis, liver cirrhosis, pregnancy.
PREGNANCY AND LACTATION
Contraindicated in pregnancy.
If it is necessary to use during the lactation period, the question of stopping breastfeeding should be solved.
APPLICATION FOR VIOLATIONS OF THE FUNCTION OF KIDNEYS
Contraindicated in acute hepatitis, cirrhosis.
